TEMPORAL STRUCTURE OF THE MUSCULAR DYSTROPHY X-LINKED MOUSE BEHAVIOUR TESTED IN OPEN FIELD

2017

In comparison with wilde type mice, animal models of Duchenne Muscolar Dystrophy (mdx mice) show a different behavioral organization with a more articulated structure of the temporal patterns. Present study sheds light, for the first time, on specific temporal features of behavior in an animal model of DMD.

Altered electrical activity in colonic smooth muscle cells from dystrophic (mdx) mice

2001

Because the colon from dystrophic (mdx) mice shows an altered motor pattern, probably due to neural disorders, our aim was to examine the electrophysiological properties of muscle cells and the functionality of nitrergic transmission in circular muscle from normal and mdx colon. Normal colonic cells (resting membrane potential [RMP] about -50 mV) showed spontaneous hyperpolarizations (inhibitory junction potentials; IJPs) and cyclic slow depolarizations were sometimes recorded. Mdx colon had a depolarized RMP (about -36 mV) and spontaneous IJPs, but the cyclic activity was never observed. In the normal colon, Nomega-nitro-L-arginine methyl ester (L-NAME) induced depolarization and abolished…

Treatment with soluble activin type IIB-receptor improves bone mass and strength in a mouse model of Duchenne muscular dystrophy

2017

Background: Inhibition of activin/myostatin pathway has emerged as a novel approach to increase muscle mass and bone strength. Duchenne muscular dystrophy (DMD) is a neuromuscular disorder that leads to progressive muscle degeneration and also high incidence of fractures. The aim of our study was to test whether inhibition of activin receptor IIB ligands with or without exercise could improve bone strength in the mdx mouse model for DMD. Methods: Thirty-two mdx mice were divided to running and non-running groups and to receive either PBS control or soluble activin type IIB-receptor (ActRIIB-Fc) once weekly for 7 weeks. Results: Treatment of mdx mice with ActRIIB-Fc resulted in significantly…

Duodenal contractile activity in dystrophic (mdx) mice: reduction of nitric oxide influence.

2003

The present study was undertaken to analyse duodenal contractility in adult dystrophic (mdx) mice. The spontaneous changes of the isometric tension and the responses of longitudinal duodenal muscle to nonadrenergic, noncholinergic (NANC) nerve stimulation and to exogenous drugs were compared between normal and mdx mice. Duodenal segments from mdx mice displayed spontaneous contractions with higher frequency than normals. N omega-nitro-L-arginine methyl ester (L-NAME) increased the frequency of contractions in normals without affecting that in mdx mice. In normals, NANC nerve stimulation elicited a transient relaxation abolished by L-NAME. In mdx mice a frank relaxation was not observed, the…

Effects of mild aerobic exercise training on the diaphragm in mdx mice

2016

Mild endurance exercise training positively affects limb skeletal muscle in the mdx mice model of Duchenne Muscular Dystrophy (DMD). However, few and controversial data are available on the effects of mild exercise training on the diaphragm of mdx mice. The diaphragm was examined in mdx and wild type mice either under sedentary conditions (mdx-SD, WT-SD) or during mild exercise training (mdx-EX, WT-EX). At baseline and after 30 and 45 days of training (5 d/wk for 6 weeks), diaphragm muscle morphology and Cx39 protein were assessed. In addition, tissue levels of the chaperonin Hsp60 were measured at the same time points in gastrocnemius, quadriceps and diaphragm in each experimental group. A…

Systemic blockade of ACVR2B ligands prevents chemotherapy-induced muscle wasting by restoring muscle protein synthesis without affecting oxidative ca…

2016

AbstractDoxorubicin is a widely used and effective chemotherapy drug. However, cardiac and skeletal muscle toxicity of doxorubicin limits its use. Inhibiting myostatin/activin signalling can prevent muscle atrophy, but its effects in chemotherapy-induced muscle wasting are unknown. In the present study we investigated the effects of doxorubicin administration alone or combined with activin receptor ligand pathway blockade by soluble activin receptor IIB (sACVR2B-Fc). Doxorubicin administration decreased body mass, muscle size and bone mineral density/content in mice. However, these effects were prevented by sACVR2B-Fc administration. Unlike in many other wasting situations, doxorubicin indu…

Effects of muscular dystrophy, exercise and blocking activin receptor IIB ligands on the unfolded protein response and oxidative stress

2016

Protein homeostasis in cells, proteostasis, is maintained through several integrated processes and pathways and its dysregulation may mediate pathology in many diseases including Duchenne muscular dystrophy (DMD). Oxidative stress, heat shock proteins, endoplasmic reticulum (ER) stress and its response, i.e. unfolded protein response (UPR), play key roles in proteostasis but their involvement in the pathology of DMD are largely unknown. Moreover, exercise and activin receptor IIB blocking are two strategies that may be beneficial to DMD muscle, but studies to examine their effects on these proteostasis pathways are lacking. Therefore, these pathways were examined in the muscle of mdx mice, …

PROTEOMIC CHANGES INDUCED BY LOW-INTENSITY ENDURANCE EXERCISE IN MDX MOUSE QUADRICEPS: CORRELATION WITH REDUCTION OF MUSCLE DEGENERATION.

2015

Previous study showed that low-intensity endurance exercise induced a significant recovery of damaged skeletal muscle in mdx mice, probably by reducing the degeneration of dystrophic muscle1. In order to explore the molecular basis of this observation, we perfomed a proteomic analysis to evaluate changes in proteins profiling of quadriceps dystrophic muscles of exercised versus sedentary mdx mice. Four protein spots were found significantly changed and were identified as three isoforms of Carbonic anhydrase 3 (CA3) and as superoxide dismutase [Cu-Zn] (SODC). Protein levels of CA3 isoforms were found significantly up-regulated in quadriceps of sedentary mdx mice (MDX-Sed) and were completely…

An attempt to enhance neurogenesis of mdx mice via aerobic exercise and myostatin inhibition

2013

Duchennen lihasdystrofia (DMD) on perinnöllinen sairaus, jonka esiintyvyys on noin 1/3600 poikavauvasta. Siihen liittyy lihasten heikkoutta, rappeutumista ja kognitiivista vajavaisuutta. Taudin aiheuttaa mutatoitunut geeni dystrophiini proteiinille. On esitetty, että kognitiivinen vajavaisuus johtuu taudin vaikutuksesta ehkäistä neurogeneesiä. Neurogeneesi on prosessi, joka jatkuvasti synnyttää uusia hermosoluja pääasiallisesti subventikulaari alueella ja hippokampuksen dentate gyruksella. Hoitoa ja parannusta tautiin tutkitaan yleensä mdx-hiirillä, joiden taudin etiologia on riittävän lähellä ihmisten tautia. Tämä tutkimus keskittyy pyrkimykseen vaikuttaa mdx-hiirten neurogeneesiin aerobis…
